Description

Principle of the Assay::The Mouse TIMP1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse TIMP1 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse TIMP1 coated on a 96-well plate. Standards and samples are pipetted into the wells and TIMP1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse TIMP1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TIMP1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: TIMP metallopeptidase inhibitor 1, also known as TIMP1, a tissue inhibitor of metalloproteinases, is a glycoprotein that is expressed from the several tissues of organisms. This protein is a member of the TIMP family. It was found to reside at Xp11.4-p11.1. This is the first growth factor found to be X-linked. The glycoprotein is a natural inhibitor of the matrix metalloproteinases (MMPs), a group of peptidases involved in degradation of the extracellular matrix. In addition to its inhibitory role against most of the known MMPs, the encoded protein is able to promote cell proliferation in a wide range of cell types, and may also have an anti-apoptotic function. What's more, TIMP1 is thought to play a regulatory role in connective tissues by forming inactive complexes with those metalloproteinases that are normally responsible for connective tissue turnover
